Bacterial DNA is a potent activator in the innate immune system through interaction with TLR9. Fragments of bacterial DNA and synthetic single-stranded oligodeoxynucleotides containing unmethylated CpG motifs (CpG ODN) found in bacterial DNA can also serve as powerful adjuvants. Different specific hexameric CpG motifs are needed to induce optimal immune enhancement between species. Use of the correct CpG motif can trigger B-cell activation and induce, either directly or indirectly, the production of Th1 and pro-inflammatory cytokines leading to the maturation and activation of APCs.
