Blocking and Incubation with Antibodies: Immunoblots Prepared with Whole-Cell Lysates and Purified Proteins (Straight Western Blotting)

(Protocol summary only for purposes of this preview site)

In this protocol, the immunoblots are prepared for detection of the proteins of interest by incubation with blocking solution to prevent nonspecific antibody binding, followed by incubation with primary and secondary antibodies. When labeled primary antibody is used in this protocol, incubation with secondary antibodies can be omitted. In brief, after the transfer of proteins and detection of the total proteins using Ponceau S dye (optional), the nitrocellulose or PVDF membrane is rinsed with ultrapure H2O. Blocking solution is applied to the membrane, and the membrane is incubated before addition of the primary antibody. The membrane is then incubated with primary antibody followed by extensive washing to remove any unbound antibody before adding secondary antibodies (standard protocol) or detection (when using labeled primary antibody).
