Labeling Antibodies Using Colloidal Gold

Colloidal goldlabeled antibodies are widely used in electron microscopy (EM). Although it is a simple procedure, it can be challenging to obtain discrete-sized gold particles without careful examination of labeled materials by EM. This difficulty has been overcome by the availability of specific sizes of immunogold. Although ultracentrifugation is still required, it is no longer necessary to vary the centrifugation speeds to separate sizes. Different-sized gold particles conjugated to secondary antibodies are also commercially available. Gold-labeled antibodies can be used for light microscopy but require additional steps (silver enhancement).
