Modifying Antigens by Arsynyl Coupling

In this technique, the protein to be used as an immunogen is modified by coupling with arsanilic acid. In the first step of the reaction, diazotized arsanilic acid is prepared, and in the second step, this reagent is coupled to the protein (Howard and Wild 1957; Amkraut et al. 1966). The diazotized arsanyl group reacts primarily with tyrosine side chains, but also binds less frequently to histidine, free amino groups, and sulfhydryl groups. Arsanilation appears to be a particularly good modification to create new sites for T-cell recognition and thus to break T-cell tolerance.
